Matching the song to him
Younger boys usually love upbeat pop with their favorite things in it. Teens tend to prefer something that sounds like their own playlists, so R&B or country may land better than a ballad. Adult sons often appreciate a quieter acoustic song that looks back.

Moments to give it
Graduation, a birthday, or the day he leaves for a new job or the military. Text him the MP3 so it goes wherever he ends up.
If he is leaving home
A son heading to college, a first job or basic training usually hears plenty of advice and not much else. A song can hold the softer part: what you will miss, what you are proud of, and that the door stays open. Keep the advice to one line at most.
If your son plays a sport or an instrument, use the specific position, number or piece. A line about jersey number 12 or the first song he learned on guitar is far more personal than 'you always loved music'.
